چکیده

This study aimed to investigate the relationship between specificity protein 1 (Sp1) expression and clinicopathological feature and prognosis of hepatocellular carcinoma (HCC). We detected Sp1 mRNA and protein expressions in 12 fresh HCC tissues by quantitative real-time polymerase chain reaction (qRT-PCR) and western blot analysis, respectively. Immunohistochemical assay was performed in 127 paraffin-embedded HCC tissues. The mRNA and protein expressions of Sp1 were significantly higher in HCC tissues than in matched adjacent normal tissues. Sp1 high expression was detected in the cytoplasm and nuclei of carcinoma cells from 68 (53.5%) patients. In addition, high expression of Sp1 was significantly correlated with HBsAg infection (P = 0.036), liver cirrhosis (P = 0.024), tumor differentiation (P = 0.010), vascular invasion (P = 0.035) and metastasis (P = 0.029). Furthermore, Kaplan-Meier analysis showed that patients with high Sp1 expression had significantly lower overall survival (OS) and disease free survival (DFS) rates, compared with patients with low Sp1 expression. Sp1 is an independent prognostic factor for HCC patients. In conclusion, Sp1 expression is significantly up-regulated in HCC and could identify a subset of HCC with aggressive clinical behavior and poor clinical outcome. Sp1 is a potential independent prognostic biomarker for HCC patients after surgery.

Analyzing the Expression Level and Associated Clinical Characteristics of miRNA-138 in Esophageal Squamous Cell Carcinoma in Iranian Patients

Background: Esophageal Squamous Cell Carcinoma (ESCC) is the seventh most common and lethal malignancy worldwide with a high incidence in Iran. Esophageal cancer has a poor prognosis and low 5-years survival rate. Despite several studies on esophageal cancer, its underlying mechanism remains unclear. Therefore, it is significant to analyze the molecular factors involved in disease for introduci...

Prognostic molecular markers in hepatocellular carcinoma (Review article

Hepatocellular carcinoma (HCC) is the 5th commonest malignancy worldwide and is the third most common cause of cancer-related death. The prevalence is different in the world. The ability to predict patients at higher risk of recurrence and with a poor prognosis would help to guide surgical and chemotherapeutic treatment according to individual risk. As understanding of hepatocarcinogenesi...

Prognostic Value of HLA-G in Malignant Liver and Pancreas Lesions

Background: Human leukocyte antigen (HLA)-G is a nonclassical HLA class I molecule with modulatory effects on NK and T cells. Because HLA-G expression is frequently detected in different solid tumors, it may be involved in tumor immune evasion. Objective: This study was designed to elucidate the prognostic value of HLA-G in hepatocellular carcinoma (HCC) and pancreatic adenocarcinoma (PADC). Th...

High Expression of Ribonucleotide Reductase Subunit M2 Correlates with Poor Prognosis of Hepatocellular Carcinoma

BACKGROUND/AIMS Ribonucleotide reductase subunit M2 (RRM2) catalyzes the production of deoxynucleotide triphosphates, which are necessary for DNA synthesis. RRM2 has been reported to play an active role in tumor progression, and elevated RRM2 levels have been correlated with poor prognosis for colorectal cancer patients. This study aimed to elucidate the prognostic significance of RRM2 protein ...

High USP22 expression indicates poor prognosis in hepatocellular carcinoma

Ubiquitin-specific protease 22 (USP22) removes ubiquitin from histones, thus regulating gene transcription. The expression frequency and expression levels of USP22 were significantly higher in hepatocellular carcinoma (HCC) than in normal liver tissues. High USP22 expression in HCC was significantly correlated with clinical stage and tumor grade. Kaplan-Meier analysis showed that elevated USP22...
